چکیده

Introduction Clinical studies have demonstrated that adoptive transfer of allogeneic Natural Killer (NK) cells can play a therapeutic role in hematological malignancies and potentially solid tumors. We established and characterized ex vivo methods to obtain murine NK1.1 cells from mouse fetal liver. These cells represent an orthologous cell type for expanded cells from human cord or placenta. Persistence, engraftment, expansion and anti-tumor efficacy of these fetal liver derived intermediate NK(FLiNK) cells were evaluated in allogeneic or syngeneic recipient mice after adoptive transfer. This study supports the possibility of clinical therapeutic application of ex vivo generated NK cells from HSC sources.

The effect of allogeneic cell therapy on tumor growth was studied in a murine model of mammary carcinoma (4T1) as an experimental model of solid tumors in humans, i.v. inoculation of 4T1 (H-2d) cells into syngeneic mice [BALB/c or (BALB/cXC57BL/6)F,J (F,) carrying the H-2d histocompatible antigens results in tumor colonies in the lungs that finally cause the death of all of the mice. Potent ex vivo expanded, human CD34+ cord blood-derived natural killer cells for cancer immunotherapy

Background Clinical studies suggest that adoptive transfer of allogeneic natural killer (NK) cells represent a promising treatment for patients with hematological malignancies and solid tumors. Celgene Cellular Therapeutics has established a cultivation process to generate human NK cells from umbilical cord blood (UCB) CD34 cells with substantial cytolytic activity against several human tumor c...

NK cell adoptive transfer combined with Ontak-mediated regulatory T cell elimination induces effective adaptive antitumor immune responses.

Previous work from our laboratory showed that hydrocortisone (HC) combined with IL-15 induces expansion of activated human NK cells. We set up an experimental tumor model to evaluate the use of adoptively transferred, HC plus IL-15 (HC/IL-15)-activated and -expanded murine NK cells in the treatment of syngeneic mice carrying established lung metastases of the CT26 transplantable tumor. We also ...

Treatment of solid tumor with autologous natural killer cells in mouse models

Cancer immunosurveillance protects the host from outgrowth of tumor cells. Natural killer (NK) cell function positively associates with reduction of cancer risk and with better survival of gastrointestinal stromal tumor patients. We expanded murine NK cells and characterized their anti-tumor activity in vitro and in vivo. Adoptive transfer of the expanded NK cells into tumor-bearing mice reduce...
